開閉自在の固体ナノポアが生体分子を識別 ―DNA塩基とアミノ酸を読み分ける次世代一分子センサへ―

大阪大学を中心とする国際共同研究チームは、化学反応によって自律的に開閉する固体ナノポアを利用し、生体分子を1分子レベルで識別できる新しいセンサ技術を開発した。従来の固体ナノポアは耐久性や半導体集積性に優れる一方、孔の大きさを動的に制御できず、DNA塩基やアミノ酸の識別が困難だった。本研究では、ナノポア内でリン酸マンガンの析出・溶解を繰り返す化学反応を利用し、「閉じる→一瞬だけ開く→再び閉じる」という開閉サイクルを自律的に実現した。この一時的な開口部を分子が通過する際の電流スパイクを機械学習で解析することで、4種類のDNA塩基分子と7種類のアミノ酸を高精度に識別し、混合試料中の組成推定にも成功した。孔が測定ごとに自動的にリセットされるため連続測定にも適しており、次世代DNAシーケンサー、タンパク質解析、医療診断、創薬、環境モニタリングなどへの応用が期待される。

Abstract

Nanopores provide a direct electrical interface to biomolecules, yet most operate as passive conduits whose sensing state is imposed externally. Here we report an autonomous solid-state nanopore in which voltage-driven precipitation and dissolution repeatedly create and erase transient conductive pathways under constant bias. This reaction cycle generates stochastic ionic spikes and converts the pore into a chemically stateful nanofluidic element whose conductance reflects recent history. As nucleotides and amino acids traverse these transient openings, they reshape the evolving pore state and produce distinct spike amplitudes, durations, and firing statistics. Learning these state-dependent fingerprints enables discrimination of four nucleotides, quantitative readout of mixed populations, and extension to seven amino acids without active gating. Our results reposition the solid-state nanopore as a reaction-programmed molecular interface that integrates sensing, memory, and signal generation within chemically dynamic nanoscale environments for molecular analytics and iontronic information processing.
