分子骨格を高速・立体的に可視化する低温原子間力顕微鏡システムを開発(Development of a Low-Temperature AFM System for Rapid Three-Dimensional Molecular Imaging)

東京大学の杉本宜昭教授らは、低温原子間力顕微鏡(AFM)による分子骨格の観察速度を従来比60倍以上に高め、世界最高速となる秒速200nmでの撮像を実現した。研究では、マイナス269℃の超高真空環境で動作するシリコン製カンチレバーとレーザー干渉方式を組み合わせた新しいAFMシステムを開発し、高感度かつ高速な力検出を可能にした。さらに、探針が試料表面の凹凸に自動追従する「トポグラフモード」を実現し、従来は困難だった立体構造や原子レベルの段差を持つ試料でも、分子骨格を壊すことなく直接観察することに世界で初めて成功した。これにより、従来30分程度を要した測定を数秒に短縮でき、表面化学反応や分子拡散などのナノスケール現象をリアルタイムで追跡する道が開かれた。本技術は、有機エレクトロニクス材料やナノデバイス開発などの基盤計測技術として幅広い応用が期待される。

Abstract

Atomic force microscopy (AFM) with a functionalized tip can resolve the chemical structure of single molecules, but its application has been limited by slow imaging speeds and the restriction to constant-height operation. Here, we overcome these limitations by operating a Si cantilever in its second resonance mode, achieving exceptional force sensitivity. This enables robust, constant-height chemical-structure imaging at speeds up to 200 nm/s—over 60 times faster than previously reported. The enhanced sensitivity also resolves molecular bonds directly in constant-frequency-shift topographic images, a mode crucial for studying nonplanar systems. Our approach significantly expands the capabilities of AFM, opening new avenues for investigating the structure of three-dimensional molecules and tracking the dynamics of on-surface chemical reactions.
